Principle

Fluorescent brightener is a kind of fluorescent dye. It has strong affinity with chitin and cellulose of fungal cell wall. Under ultraviolet light (wavelength: 340-380nm), it will emit bright blue fluorescence. Therefore, the morphology was observed by a fluorescence microscope to qualitatively detect the fungus.

Sample requirement

  • 1. Fresh or frozen tissue sections.
  • 2. Samples of all kinds of dander, armor, hair, sputum, body fluids, and vaginal secretions.

Test procedure

  • 1. Pick up the sample and make it into a smear, fully fix after natural drying, or dewax the tissue section to water step in a routine manner.
  • 2. Place the specimen in a horizontal position, add a drop of fungal fluorescent staining solution on the specimen, and cover it with a cover slip to fully integrate it with the specimen.
  • 3. The staining was kept for 2 minutes, and excess dye was removed with a filter paper and observed under a fluorescence microscope (wavelength: 340~380 nm).

Result interpretation

  • The mycelia and spores of the fungus were all bright blue, and the structure was clear and distinct, other bacteria and tissues showed weak blue fluorescence and the background was blue-black. However, the diagnosis of fungal infection should be based on clinical experience.

Method limitation

  • Only used for morphological staining to observe.

Precautions

  • 1. After the cover glass is pressed, use the filter paper to remove excess dye, so that the effect under the microscope can be clearer, and at the same time avoid the liquid from overflowing and causing pollution.
  • 2. Please read the smear as soon as possible after staining, to avoid liquid dryness affect the observation.
  • 3. Please use a fluorescent filter with a wavelength range of 340nm~380nm in the darkroom environment. Otherwise it could cause false negative results.
  • 4. For kit storage, avoid exposure to extreme high or low temperature and sunlight.
  • 5. Each time reagent after use, please quickly cover, to avoid evaporation and affect the result.
  • 6. This kit must be used by professionals and the interpretation of results.
  • 7. Should carefully read the instruction manual before using. Can only be used before expiration date, and good personal hygiene protection.
  • 8. After use, disposal of waste should comply with the hospital or the environmental protection department requirements.
  • 9. Production date and lot number and expiration date showed on the package.
