Preparation of Naringenin-Loaded β-Casein Micelles

Bovine milk protein β-casein (β-CN) is an intrinsically disordered amphiphilic protein that can self-assemble into micelles. Naringenin, a major hydrophobic flavanone found in grapefruit, exhibits various beneficial biological effects such as antioxidant, anticancer, and anti-inflammatory activities. This study demonstrates that naringenin can be encapsulated in β-CN micelles.
Preparation of naringenin-loaded β-CN micelles: β-CN solutions of different concentrations (1, 2, 3, 4, and 5 mg/mL) were prepared in 20 mM sodium phosphate buffer (pH 7.0). Naringenin stock solution (0.168 M in ethanol) was added to the protein solution under magnetic stirring for 2 hours, resulting in a final naringenin/β-CN molar ratio of 12:1. Centrifugation (13,000 rpm, 15 minutes) did not cause naringenin precipitation, indicating complete dissolution of naringenin. The supernatant was stored at 4 °C for further analysis.
