N-propargyl caffeamide (PACA) Enhances Conversion of proNGF to NGF in Midbrain Tissues

To determine the effect of PACA on NGF and proNGF protein levels in mouse brain, NGF and proNGF protein levels were determined by Western blotting using specific antibodies. In mice treated with MPTP, NGF protein levels in SNpc were reduced. PACA effectively restores NGF protein levels to resist the neurotoxicity of MPTP. MPTP greatly increased the accumulation of proNGF, while PACA reduced proNGF protein levels in the midbrain of mice treated with MPTP.
In order to verify the effect of PACA on NGF and proNGF protein levels, we further measured NGF levels through immunofluorescent staining. Midbrain tissue was recovered from mice after treatment with a combination of vehicle, MPTP, MPTP and PACA. MPTP almost eliminated binding of anti-NGF antibodies, while PACA restored retention of anti-NGF antibodies in the midbrain of MPTP-treated mice. Based on immunostaining of NGF and GFAP in the substantia nigra, a dose of 15 mg/kg/day PACA increased NGF levels not only in astrocytes, but also in other cell types. These results suggest that PACA increases the conversion of proNGF to NGF in midbrain tissue.

N-Propargyl Caffeate Amide (PACA) Enhances NGF-Induced Neuritogenesis

To evaluate the neuritogenic activity of PACA, PC12 cells were treated with NGF (2 ng/mL) alone or in combination with PACA at concentrations of 5, 10, and 20 μM for 72 hours. Neurite outgrowth was then analyzed by staining the cells with a neurite outgrowth staining kit and examining them under a fluorescence microscope to identify neurites longer than 20 μm. Compared to NGF alone, PACA significantly enhanced NGF-induced neuritogenesis in a concentration-dependent manner. Notably, PACA alone at a concentration of 20 μM did not induce neuritogenesis. Interestingly, the combination of PACA (20 μM) and NGF (2 ng/mL) resulted in an approximately six-fold increase in the number of neurite-bearing cells compared to NGF alone. After treating PC12 cells with PACA and NGF, either alone or in combination, the expression of neuronal biomarkers such as β3-tubulin, MAP2, and GAP-43 was analyzed by Western blotting. PACA significantly enhanced NGF's activity in inducing these neuronal markers. In conclusion, these results suggest that PACA can potentiate NGF's neurogenic activity in terms of neurite extension.

N-Propargyl Caffeamide (PACA) Protects the Heart from Ischemia-Induced Cardiac Injury

To evaluate the cardioprotective effects of PACA in vivo, myocardial infarction was induced in rats by ligating the left anterior descending (LAD) artery. PACA was administered via intraperitoneal injection 24 hours after arterial ligation. According to TTC staining, PACA reduced the myocardial infarction area from 34.5% to 15.0% by the third day. H&E staining revealed the destruction of myofibril structure and the accumulation of neutrophil/macrophage clusters in the ischemic myocardium. PACA at a dose of 15 mg/kg not only preserved myofibril structure but also reduced the recruitment of inflammatory cells and myolysis.
