Principle

After tissue fixed by containing chromium salt fixative, with Giemsa stain, the chromaffin cell granule was stained green-yellow.

Sample requirement

  • Fresh tissue was immediately placed in Regaud's solution for 2 to 4 days, and the new solution was changed once a day, and then fixed with 3% potassium dichromate aqueous solution for 1 day, rinsed with running water for 24 hours, and dehydrated by conventional dehydration. The tissue section must be fully fixed.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water, about 30 minutes.
  • 2. Placed into Giemsa diluent (A Solution: B solution = 1:20) dip stain 18~24 hours, washed with distilled water slightly and then dried with filter paper.
  • 3. Differentiated few seconds with n-butanol (Purchase separately), then dried with filter paper.
  • 4. Xylene transparent, sealed with neutral resin.

Precautions

  • 1. Regaud's fixative preparation: mix 3% potassium dichromate 80ml and 20ml concentrated formaldehyde solution before use, after mixed for 24 hours it will be gradually lapsed.
  • 2. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 3. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
