Principle

This method mainly uses acid fuchsin and picric acid to double dye the collagen fiber and the muscle fiber respectively. The bitter acid has a small molecular weight, can penetrate muscle fibers, glial cells and red blood cells with dense tissue structure combining with bitter acid were stained in yellow, The acid fuchsin has a medium to large molecular weight, and can only penetrate collagen fibers with looser tissue structure. combining with acid fuchsin were stained in to red to pink.

Sample requirement

  • The tissue section must be fully fixed.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water.
  • 2. Add Weigert's Iron Hematoxylin Solution A, and then add Weigert's Iron Hematoxylin Solution B mixed with it (A: B = 1:1), staining for 5~10 minutes, slightly washed.
  • 3. Rapidly differentiate with 1% hydrochloric acid alcohol and wash for several minutes.
  • 4. Staining with Van Gieson stain (abbreviation: VG) for 1~2 minutes.
  • 5. After pouring Van Gieson stain, directly differentiated with 95% alcohol for several seconds.
  • 6. Conventional dehydration and transparent, sealed with neutral resin.

Precautions

  • 1. Weigert’s Iron Hematoxylin is divided into two liquids A and B. Should mix with two liquids prior to use. It is not suitable to pre-mix, otherwise it will easily oxidize and precipitate then gradually lose the staining power.
  • 2. Differentiate tissue quickly in 95% ethanol after Van Gieson stain. Otherwise it is easily discolored excessively.
  • 3. Because acid fuchsin is soluble in water, after staining in VG solution do not rinse in water. Instead, add 95% ethanol directly to differentiate and then dehydrate in absolute ethanol. Otherwise, the colors from the VG stain will weaken or discolor.
  • 4. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 5. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 6. The production date, production batch number and expiration date are shown in the outer packaging.
