Principle

Aldehyde fuchsin has a strong affinity regarding to the containing sulfate group of mucopolysaccharide material, inside Mast cell granules has containing nucleic acid and carboxyl group of heparins, so easy and aldehyde fuchsin combined into complexes and coloring.

Sample requirement

  • The tissue should be fresh and quickly fixed, and its particles destroyed by the long-term storage of the tissue.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water.
  • 2. Add 75% ethanol wash slightly.
  • 3. Add Aldehyde fuchsin solution staining for 10~15 minutes, 75% ethanol differentiated and wash away excess stain solution, slightly water washed.
  • 4. Orange G solution drop staining about 3~5 seconds, slightly water washed.
  • 5. Conventional dehydration and transparent, sealed with neutral resin.

Precautions

  • 1. Mast cells staining require fresh tissue and quickly fixed.
