Principle

Mast cell particles contain heparin and histamine etc. These are sulfuric acid esters, are heterochromatic, so with the metachromatic dye toluidine blue staining, it can be appeared heterochromatic purplish-red.

Sample requirement

The tissue should be fresh and quickly fixed, and its particles destroyed by the long-term storage of the tissue.

Test procedure

1. The slices are routinely dewaxed to water and washed once with distilled water.
2. Toluidine blue solution staining for 20～30 minutes, slightly water washed.
3. Differentiation of glacial acetic acid solution, until cell nuclear and particle clearly (Control under a microscope), slightly water washed.
4. Dry with cold air (Not through alcohol), transparent by xylene then sealed with neutral resin.

Result interpretation

Mast cell granules	Purplish-red
Cell nuclear	Blue

Precautions

1. Mast cells staining require fresh tissue and quickly fixed.
2. Due to ethanol easy mast cell particles restore the orthochromatic light blue, so after staining, the section with cold air dried, do not use ethanol dehydration.
3. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
4. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
5. The production date, production batch number and expiration date are shown in the outer packaging.
