Principle

Ammoniacal silver solution was adsorbed by tissue, and combined with the protein within the tissue, via formaldehyde reduction into black metallic silver deposited on the surface and inside of tissue. After toning with gold chloride, and then using sodium thiosulfate solution wash the unreduced silver, thus the reticulum within the tissue clearly displayed.

Sample requirement

  • The tissue section thickness about 5μm, and must be fully fixed.

Test procedure

  • 1. The slices are routinely dewaxed to water.
  • 2. Potassium permanganate solution oxidized for 3 minutes. Wash in running tap water.
  • 3. Bleach with oxalic acid solution for 1~2 minutes until the tissue is colorless, rinse with running water.
  • 4. Ferric ammonium sulfate solution for 3 minutes. Rinse in deionized water 10 seconds.
  • 5. Ammoniacal silver solution for 5 minutes (Tightly cap the bottle of Ammoniacal silver solution immediately to avoid ammonia volatilization then lead to failure), Quickly rinse in deionized water 5~10 seconds.
  • 6. Reduction of formaldehyde solution for 30 seconds, Wash in running tap water.
  • 7. Toning with Gold chloride solution for 1 minute, control under the microscope if necessary, Wash in running tap water.
  • 8. Sodium thiosulphate solution for 2 minutes, wash in running tap water.
  • 9. Conventional dehydration and transparent, sealed with neutral resin.

Precautions

  • 1. Ammoniacal silver solution must be away from light and at low temperature storage.
  • 2. All containers used in this method, should be pre-soaked with cleaning solution and rinse. After use, containers should be immersed with cleaning solution.
  • 3. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 4. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 5. The production date, production batch number and expiration date are shown in the outer packaging.
