Principle

Diff-Quik Stain is prepared by the World Health Organization (WHO) recommended staining method. Like Wright Stain, it is improved by using the principle of Romanowsky Stain. And the staining results are similar. However, Diff-Quik Stain takes a relatively short time and the staining can be completed in about 1 minute. It is more suitable for batch dipping, which saves staining time, clear background and no sediment.

Sample requirement

  • 1. Blood cell smear staining: requires fresh whole blood or EDTA·K2 anticoagulation.
  • 2. Vaginal secretions (gynecological leucorrhea) smear staining: fresh specimens need to be fixed with flame or alcohol as soon as possible to avoid cell deformation after leaving the human smear.

Test method

  • 1. Take dip-staining method, and the staining cylinders need to be sealed, please take a trial staining before staining, because each operator has different habits of staining effect. Please extend or shorten the time to adjust to the most suitable effect of the morphological observer.
  • 2. Allow the smears to air dry.
  • 3. The smear is immersed in the Diff-Quik Fix for 20~30 seconds, and erect the slide on the filter paper to remove excess solution.
  • 4. The smear is immersed in the Diff-Quik I for 5 seconds, and erect the slide on the filter paper to remove excess solution.
  • 5. The smear is immersed in the Diff-Quik II for 15~20 seconds, and remove the excess staining solution by being immersed in deionized water.
  • 6. Smear dried, and then observed with the microscope.

Result interpretation

  • RBC appear light red, the cytoplasmic granules in WBC is clear, and various cells display their distinctive colors. Nucleus is purple-red, and the nuclear chromatin structure is clear.

Precautions

  • 1. During staining process, the staining cylinder must be sealed after stained, otherwise it will be easily volatilized  and cause poor staining.
  • 2. Shake the reagent before pouring it into the dye vat. After using, please cover it quickly to avoid evaporation.
  • 3. When storing reagents, try to avoid high and low temperature environment and direct sunlight.
  • 4. When the room temperature is too low in winter, the staining time should be extended appropriately.
  • 5. This reagent should be used by professionals.
  • 6. Read the instruction manual carefully before use, use within the validity period, and do personal hygiene protection.
  • 7. After use, dispose the wastes according to the requirements of the hospital or environmental protection department.
  • 8. The production date, production batch number and expiration date on the package.effectiveness of this kit.
