Principle

Carmine can bind mordant aluminum to form a complex, which in turn binds to the acid group of mucus and becomes red.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water.
  • 2. Place in hematoxylin solution (Mayer) for 10 minutes and rinse briefly with running water.
  • 3. Blue in Scotts tap water substitute solution for 1 minute and rinse briefly with running water.
  • 4. Take a clean container, add 1ml Mucicamine solution and 3ml distilled water, mix it into a dilute the red liquid and staining 20~30 minutes.
  • 5. Rinse briefly with running water.
  • 6. Counterstain in Tartrazine solution for 1~3 minutes after blotting up with filter paper.
  • 7. Rinse briefly with running water.
  • 8. Conventional dehydration and transparent, sealed with neutral resin.

Precautions

  • 1. When hematoxylin is used to stain the nucleus, Ehrlich hematoxylin should be avoided because it can lightly stain the mucus and obstructs mucous coloring.
  • 2. Store Mucicamine solution at 4℃. Put the bottle cap back into the refrigerator after each use, otherwise it will affect its service life.
  • 3. Mucicamine diluent should be prepared prior to use, if placed it at room temperature or refrigerator on the day can also be used, but the effect will be slightly reduced.
  • 4. Tartrazine solution counterfeit dye cannot be excessive, otherwise it will cover the red mucus, but also, can leave out the staining step.
  • 5. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 6. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 7. The production date, production batch number and expiration date are shown in the outer packaging.
