Principle

Bouin's solution is one of the most used mixed fixatives in histochemistry. It is made up of a mixture of picric acid saturated solution, formaldehyde and glacial acetic acid in a certain ratio. The glacial acetic acid in the fixing solution has an expansion effect on tissues such as collagen fibers, which can offset the contraction of formaldehyde to tissues and achieve an excellent fixed level of tissue fixation. Bouin's solution can be used as a fixative or as a staining solution, but it cannot be used as a preservation solution. It does not extend the shelf life of the tissue. The maximum fixed time cannot exceed 48 hours.

Sample requirement

  • Fresh tissue or fully dewaxed paraffin sections.

Test procedure

  • 1. Tissue pre-fixation: After fresh tissue repair, placed it in Bouin's solution for 10~24 hours, and rinsed it overnight with running water, or directly placed in 70% ethanol without rinsing, and conventionally dehydrated and embedded.
  • 2. Tissue supplemental fixation: If the tissue has been fixed with 10% formaldehyde solution, can be dewaxed into water after the slice, and then put it into Bouin's solution for 10~24 hours or into 37℃ incubator for 1~2 hours, rinsed these sections with running water until the yellow color disappears then stained .

  • 1. This product is mainly used for the fixation of tissues or cells, and belongs to the reagents for pre-staining of specimens.
  • 2. This product has a wide range of applications, especially suitable for specimens and embryo specimens rich in connective tissue, and can also be used for the fixation of plant tissues.
  • 3. Because this product contains bitter acid, it will dissolve the fire-cotton glue, so if it is used for the embedding of fire-cotton glue, it must be treated with elution of picric acid.
  • 4. After each reagent is used, please quickly seal and store it to avoid evaporation and affect the effect.
  • 5. This product should be used by professionals and the interpretation of the results.
