Principle

Hepatolenticular degeneration, also known as Wilson's disease, is an autosomal recessive genetic disorder of copper metabolism disorders. Due to histologically poor liver disease specificity of Wilson's disease, so copper staining of liver tissue is very important for the identification and diagnosis of Wilson's disease. Rhodanine is a two-position chelator. It has a strong affinity for copper and can be used to detect the precipitation of copper particles.

Sample requirement

  • The tissue section must be fully fixed.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water. Slightly washed with 95% ethanol.
  • 2. The sections were immersed in rhodamine staining solution, sealed with a lid, and stained in a 37℃ incubator for 24 hours or in a 60℃water bath for 3 hours.
  • 3. Remove the section, slightly rinse with 95% ethanol, and rinse with running water for 10 seconds.
  • 4. Counter-stain with hematoxylin (Mayer) solution for 2 minutes and rinse under running water for 3 minutes.
  • 5. Conventional dehydration and transparent, sealed with neutral resin.

Precautions

  • 1. When the sections are immersed in the rhodamine solution, they should be sealed to prevent volatilization of the staining solution, at the same time, the amount of the staining solution should be sufficient, the staining temperature should be constant, and the staining time should be sufficient.
  • 2. Due to the long staining time, attention should be paid to the prevention of tissue removal in the staining process. The prevention method is to pay attention to the cleanliness of the slide during the tissue production process, and the flat sheet is free of wrinkles, the temperature and time of the slide heating are enough.
  • 3. Rhodamine solution can be reused 4~5 times.
  • 4. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 5. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 6. The production date, production batch number and expiration date are shown in the outer packaging.
