Principle

Periodic acid is an oxidant which can destroy carbon bond of polyrose. First, tissues are oxidized in periodic acid solution to break open the carbon bond of glycol group or ammonia hydroxyl in tissue polyose, which results in the formation of aldehyde compound. Subsequently, the free aldehyde exposed will react with colorless fuchsin to form a new red- fuchsia compound.

Sample requirement

  • The tissue section must be fully fixed.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water.
  • 2. Rinse in distilled water for 1~2 minutes.
  • 3. Oxidized sections in Periodic acid solution for 10 minutes.
  • 4. Rinse thoroughly in distilled water and dry sections with absorbent paper.
  • 5. Stain in schiff's reagent for 10~15 minutes.
  • 6. Rinse thoroughly in water and dry sections with absorbent paper.
  • 7. Stain nucleus with Hematoxylin solution (Mayer) for 2~4 minutes.
  • 8. Rinse with running water for 3~5 minutes, remove excess water.
  • 9. Conventional dehydration and transparent, sealed with neutral resin.

Precautions

  • 1. Mayer is recommended to store at room temperature for better coloring effect.
  • 2. Schiff's reagent was taken out of the room and recovered to room temperature within half an hour before use. It cannot be used when the unstained reagent appears light red.
  • 3. If the kit is stored at 2~8℃, the reagent will gradually turn light red during the dyeing process, which is a normal phenomenon.
  • 4. The staining time in colorless fuchsin solution will depend on the room temperature. In summer, 10 minutes is typically enough, in winter, however, it should be extended up to 20 minutes.
  • 5. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 6. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 7. The production date, production batch number and expiration date are shown in the outer packaging.
