Principle

This is a ferrous ion adsorption method, melanin can intake the ferrous ions, that is ferrous ions adsorbed on melanin, formed melanin ferrous complex, then potassium ferricyanide combined with ferrous ion. Generated the iron ferrous cyanide in the acidic environment that is Turnbull's Blue reaction.

Sample requirement

  • The tissue section must be fully fixed.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water.
  • 2. Add Ferrous sulfate solution for 1 hour.
  • 3. Distilled water washed 3 times, each about 3~5 minutes.
  • 4. Add potassium ferricyanide acetic acid solution for 30 minutes.
  • 5. Glacial acetic acid solution wash slightly 2~3 seconds, dry it.
  • 6. Staining with the Van Gieson stain about 1 minute.
  • 7. After pouring Van Gieson stain, directly differentiated with 95% alcohol for several seconds.
  • 8. Conventional dehydration and transparent, sealed with neutral resin.

Precautions

  • 1. This method is fixed with formaldehyde or Carney solution is better, should not use chromate fixative.
  • 2. The hemosiderin sometimes was green reaction, to note the difference, necessary to do the iron stain to proof.
  • 3. After staining with Van Gieson solution, it should be rapid when it is differentiated by 95% alcohol, otherwise it will be easily discolored.
  • 4. Because acid fuchsin is soluble in water, Van Gieson solution cannot be washed after washing, otherwise the color stained by Van Gieson solution will be weakened or even eluted.
  • 5. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 6. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 7. The production date, production batch number and expiration date are shown in the outer packaging.
