Principle

It is a metal replacement method. When silver nitrate solution acting on the insoluble calcium salts of histological section, calcium salt is converted to silver. Silver salt will be reduced to the black metallic silver because of the effect of light (such as sunlight or ultraviolet light).

Sample requirement

  • The tissue section must be fully fixed.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water.
  • 2. Drop silver nitrate solution on the tissue section or dip it in silver nitrate solution, then place them in strong sunlight for 15~60 minutes, It takes only 5~10 minutes if exposed to UV light, then rinse the tissue section with distilled water for 3 minutes.
  • 3. Place it in sodium thiosulfate solution for 2 minutes, then rinse the tissue section with running water for 5minutes.
  • 4. Place it in Hematoxylin (Harris) (Purchase separately) for 4~6 minutes, slightly rinse the tissue section with water.
  • 5. Differentiate in 1% hydrochloride alcohol solution (Purchase separately) for few seconds, rinse the tissue section with running water.
  • 6. Stain the tissue section in Eosin stain (Aqueous) (Purchase separately) for a minute, slightly rinse the tissue section with water.
  • 7. Conventional dehydration and transparent, sealed with neutral resin.

Result interpretation

  • Calcium salt appears brown black to deep black.

Precautions

  • 1. Must use neutral buffered formaldehyde solution to fix calcium salt , rather than acidic fixative because acid will dissolve portion of calcium salt.
  • 2. Could counterstain it with Van Gieson solution (Purchase separately) instead of H-E solution.
  • 3. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 4. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 5. The production date, production batch number and expiration date are shown in the outer packaging.
