Principle

The nucleus in the cell is composed of acidic substances, it has a strong affinity with the basic dye, and the cytoplasm is the opposite, it contains alkaline substances and has a strong affinity with the acid dye. Papanicolaou staining solution using this feature on the cell coloring, and the cellular structure can be clearly stained. Cytoplasm is translucent bright; a variety of particles and the nucleus chromatin are clear. So, it's easier to find abnormal cells. Papanicolaou staining can reflect the morphological changes of cell after inflammatory stimulation and cancer. It has a great significance of detection and diagnosis of some lesions and tumors in the early.

Result interpretation

EpitheliaNuclear is indigo and nucleolus is red, keratinized cells in cytoplasm appear pink, full-keratinized cells appear orange, cells prior to keratinization are sky-blue or light green.

Precautions

  • 1. It is normal to produce a layer of oxide film on the surface of hematoxylin stain (Harris) and a little precipitation of aluminum sulfate crystals on the bottom. Before use, the oxide film should be removed and filtered regularly. Dilute lithium carbonate solution (a package of lithium carbonate powder is dissolved in 1000ml distilled water) is used for blueing.
  • 2. Hematoxylin stain (Harris) is not easy to stain when the temperature is low, and the staining time can be appropriately extended.
  • 3. The color separation is to use 1% hydrochloric acid alcohol solution to wash away the excessive hematoxylin absorbed by the cells, so that the nuclear and cytoplasmic contrast is distinct. The differentiation time should not be too long, otherwise the nucleus will be light stained.
  • 4. Orange G remained on the slide should be removed by washing in 95% ethanol. Otherwise, EA36 or EA50 Solution staining will be affected.
  • 5. Do not use the reagents beyond the stated expiration date. For storage, avoid exposure to extreme high or low temperature and sunlight.
  • 6. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 7. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 8. The production date, production batch number and expiration date are shown in the outer packaging.
