Principle

Toluidine blue has strong heterochromaticity. When acidic mucopolysaccharides are present in the cells, especially sulfur-containing acidic mucous substances, the color of toluidine blue can be changed from blue to red. The basophilic particles in basophils are mainly acid mucopolysaccharides and belong to sulfate esters, which are heterochromatic. Therefore, staining with metachromatic dye toluidine blue can make them appear heterochromatic red-purple.

Sample requirement

  • Fresh bone marrow smear should be fixed in time.

Test procedure

  • 1. After the bone marrow smear is naturally dried, add toluidine blue staining solution for 5 minutes (do not pour out the staining solution or let the staining solution dry).
  • 2. Then add the same amount of distilled water to mix with the staining solution, and mix thoroughly with a rubber pipette bulb, and staining for 15 minutes.
  • 3. Wash it gently with water, let it dry naturally, and then check it under a microscope.

Result interpretation

The basophilic granules are red, light pink or purple, and the background cells are light blue.

  • Reference range:
  • 1. The basophils of basophilic leukemia are strongly positive.
  • 2. When chronic myelogenous leukemia has acute basophilic changes, it is specific to the identification of basophils.

Precautions

  • 1. In order to confirm the reliability of the staining effect, a patient's blood smear containing basophils can be selected as a control during each staining.
  • 2. The product should be used and the interpretation of the results should be made by professionals.
  • 3. Read the instructions before use, use within the validity period, and keep a personal hygiene protection.
  • 4. After use, the disposal of waste should according to hospital or environmental protection department requires.
  • 5. The production date, production lot number and expiry date are in the packaging.
