Principle

Capsule is a characteristic mucous substance developed external to but contagious with the cell wall during bacteria's life cycle. The capsule can protect bacteria from phagocytosis and subsequent digestion by themhost, thus increasing their infectivity. Capsules can only be stained with certain special stains. Identification of capsules is very important for bacteria grouping and differentiation.

Sample requirement

  • Pick colonies and make smear, ethanol-fixed after natural drying.

Test procedure

  • 1. Add crystal violet solution, then heat on flame and keep staining solution steaming for about 1 minute.
  • 2. Wash dyes on the smear with copper sulfate solution (avoid washing with water).
  • 3. Dry with absorbent paper and observe under oil immersion lens.

Method limitation

  • Only use for morphological staining to observe.

Precautions

  • 1. As water represents 90% of the capsule contents, do not fix samples by heating or capsules will shrink.
  • 2. After staining, samples must be washed with copper sulfate solution instead of water.
  • 3. This kit must be used by professionals and the interpretation of results.
  • 4. Should carefully read the instruction manual before using. Can only be used before expiration date, and keep a good personal hygiene protection.
  • 5. After use, disposal of waste should comply with the hospital or the environmental protection department requirements.
  • 6. Tightly cap the reagent bottle immediately after use to avoid evaporation.
  • 7. Production lot number and expiration date are on the package.
