Principle

The principle of this method is not well-known. Hypothesis includes that hydrogen bond forms between certain parts of elastic fiber and phenolic group of resorcinol in Elastin Stain, consequently the elastic fiber is stained blue-black. VG stain was used for contrast stain.

Sample requirement

  • The tissue section must be fully fixed.

Test procedure

  • 1. The slices are routinely dewaxed to water and washed once with distilled water.
  • 2. Oxidized in potassium permanganate solution for 5 minutes. Rinse in water.
  • 3. Bleach in oxalic acid solution for 2~3 minutes. Rinse in water.
  • 4. Rinse slightly in 95% ethanol and stain in Elastin stain for 8~24 hours (stamped, avoid volatile dye).
  • 5. Differentiate in 95% ethanol or 1% hydrochloric acid alcohol (if necessary, under control of the microscope).
  • 6. Rinse thoroughly in water for 3~5 minutes.
  • 7. Counterstain in Van Gieson stain for 1 minute (without washing).
  • 8. Differentiate quickly in 95% ethanol for several seconds.
  • 9. Conventional dehydration and transparent, sealed with neutral resin.

Precautions

  • 1. Differentiate tissue directly in 95% ethanol after Elastin stain, without rinsing in water, to avoid hard-to-differentiate.
  • 2. As Elastin stain is easy volatile and takes a long time to complete the staining, please using dip stain in sealed container.
  • 3. After staining in Van Gieson stain, do not rinse in water. Otherwise, the colors stained with Van Gieson stain will weaken or even discolor.
  • 4. After the Elastin stain is unsealed, it should be placed in a cool, dark, airtight container, it can be reused until the stain solution fails.
  • 5. Elastin stain can also be used 37℃ water bath (capped) staining 30~60 minutes to accelerate staining, but under the microscope control to prevent staining too deep, The stain solution after heating and used, the number of reusables will be reduced.
  • 6. This reagent should be used by professionals and interpretation of the results. Read the instruction manual carefully before use, use it within the validity period, and do personal hygiene protection.
  • 7. Dispose of waste according to the requirements of the hospital or environmental protection department after use.
  • 8. The production date, production batch number and expiration date are shown in the outer packaging.
